I want to extract RNA from fish blood for making cDNA. What would be suitable temperature for preserving the blood sample in case of delayed extraction of RNA from the blood? Does RNA degrade if I want to extract it later?
Hi Muhammad, RNA degrades continually no matter what you do. If you are in the field collecting fish blood with limited resources, I think the best you can do is to have a dry ice container to store~1ml of blood samples.
If you cant get dry ice, then use an ice chest. If not, then prepare some tubes with Trizol to collect the blood.
I think you can use RNA later (Sigma or any other company) to collect as well as store theRNA samples. Generally, we use to store RNA samples at -80 degree C.
If you're not able to flash freeze the samples and store at -80C, I would recommend using RNALater as Gunajit described. After storage in RNA Later, there's a kit you can use that's made for isolating RNA from blood (link attached).
Just FYI - the yield is really low (Average yields of RNA are between 2–4 µg/0.5 mL of whole blood). Depending on the fish you're using, it may not be possible to get that amount of whole blood.
