Hi, did you check if other TNF-dependent genes were upregulated in the same endothelial cells? This may give you a hint if your TNF is OK ... other thoughts ... what about stimulation time (I think mRNA response is very quick and transient ... max. about 4 hours)? .... how many units/ml TNF did you use?

I ve tried with 2, 5,10ng.ml (fianl concentration) and the time course was 10 and 24 hours. TNF is OK because I incubated HAEC with tha same TNF and there was alteration in expression of adhesion molecules (ICAM VCAM). I will change the stimulation time. Thank you

Most people take 2-50 ng/ml TNF ... may be your TNF concentration ist a little bit to low and the incubation time to long ... I would try 5, 15, 30 ng/ml and shorter times ... e.g., 3, 6, 10 hours. Good luck!
