I made the vitamin solution acc. to Wolfe as follows: I made stock solutions of these compounds at 10 mg per mL, then for each compound the corresponding volume was diluted in 50 mL, and the mixture was filter-sterilized, by a 0.2 um syringe filter into sterile water to complete 1 L.

  • 10mg Pyridoxine hydrochloride
  • 5.0mg Thiamine-HCl
  • 5.0mg Riboflavin
  • 5.0mg Nicotinic acid
  • 5.0mg Calcium D-(+)-pantothenate
  • 5.0mg p-Aminobenzoic acid
  • 5.0mg Thioctic acid
  • 2.0mg Biotin
  • 2.0mg Folic Acid
  • 0.1mg Vitamin B12

The 50 mL solution containing all components looks translucent and without precipitated compounds, but following filtration the syringe filter looks orange-ish and the 1 L solution has a deep green fluorescent color. Is this normal? Should I be concerned that something orange (either the folic acid or the riboflavin) did not pass the filter? It is the first time I, or someone in this lab makes said solution so it is a bit puzzling that there could be a problem with filtration.
