Hello all! I am currently looking at either using Exo-CIP rapid clean up or ExoSAP-IT (non-express). I was hoping to get some people's opinions on these two products. It looks like Exo-CIP express is cheaper and faster, but I'm not sure if it is as reliable as ExoSAP-IT. I am using the PCR products for Sanger sequencing. Any input would be greatly appreciated.
I have used exo-sap for thousands of samples and it is excellent. It has one fault which will also apply to exo-sip. The exo enzyme does not degrade double stranded dna so if you have one primer that has either a loop caused by CC GG binding or by a G quadruplex then it will only partially destroy that primer because the neck of the loop is DS dna. If you then add the other primer for sequencing yu will get 2 sequences...one from each end and the result will be a mess. It does not happen often but be aware that it can happen
