Has anyone quantified protein from their Extra-cellular Vesicle isolate using a microBCA kit ? I am not picking up concentrations (for BSA standards) anywhere near the kit's sensitivity (= 0.02 ug/ul). The smallest concentration I can read is 0.125 ug/ul on a microplate reader.
Any help would be greatly appreciated.

I have measured the protein concentration of exosomes using NanoDrop and the smallest concentration I got, was about 0.06 ug/ul. I haven't used a microplate reader. So microBCA kit worked for me!
I used to do the BCA tube assay with my lowest standard concentration as 0.05mg/ml. This could be detected consistently and accurately on the Nanodrop (nice linear standard curve).

Actually, I'm trying the same with microBCA and I still have some problems with the protein estimation.
Do you have some tip or any optimized protocol to share or to reference?
