I have heard of people using ethanol instead of methanol for their western blot transfers because it is more environmentally friendly. However, they were doing wet transfers, and we are using a Biorad semi-dry transfer system. Does anyone know if ethanol also works for semi-dry transfers. If so, how much? 20%?
Its absolutely no problem to use ethanol instead of methanol in my experience. We used actually both, depending on the availability in the lab. But: If you start with ethanol you have to go through the process with it and vice versa.
Ethanol is also toxic to the environment (although not as much as methanol).
We use ethanol instead of methanol (same percentage) for our Biorad semidry system. It works well for most proteins, but I prefer wet transfer, because the risk of blotting artifacts by bubbles e.g. is lower and the risk of blotting your proteins through the membran.

HI, I do not use ethanol and methanol in transfer buffer with biorabd semidry transfer apparatus and it is giving very good transfer efficiency. I never had a problem of transfer at all and had good results in western.
I've never used ethanol for WB but if it is possible, it will be great! Let we Know!
