I assume you know the principal difference between in vivo and in vitro. There can be differences. for example in hormone physiology in vivo can be more sensitive than in vivo. In vivo a smaller dose might be enough for a response than in vitro. In vitro you might be measuring how much of a hormone you need for a full response, while in vivo, a smaller amount give you a full response.
to measure detoxifying enzyme activity you can go in two ways; 1. you can collect your insects and do assays separately 2. you can assay them based on the LC50 or LC30 or less! treat your insects with these concentrations (or others, depends on what do you need!) and then assay their enzymes! in this method you can find out the effect of insecticides on your insects enzymes!
Different pesticides acting enzyme inhibitors showed different effects in vivo and in vitro. They also demonstrate variability among different pest insects depending on physiology and cuticle penetration of insects concerned. Some pesticides can readily absorbed through cuticle of insects other or not so easily. some pesticides works swiftly through skin than through ingestion. pesticides are specific in action e.g. stomach or contact chemicals.
