I have a student interested in trying embryo rescue after different species pollination. Can anyone suggest some papers or give general tips for successful tissue culture, media, hormones, dissection techniques
Your question is typical. In general main problem in tissue culture is contamination and development of phenolic compounds. Time factor and composition of media are very important. Once you excise the required parts (ex-plants) immediately transfer to nutrient media in sterile conditions so that you can avoid development of phenolic compounds. Composition of media varies basing on the development stage, prepare (use) according to that. You mentioned that you work on different species, no idea what species you work, vast literature is available on variety of plant species. Search according to that. You get lot of information.
