I have a practical problem. The department where I am employed does not posses a Tecan reader, therefore I have to drive 15 min to get to a sufficient scanning equipment.
The question is:
“Which ELISA step would it be least detrimental for the analysis and most practical for transportation?”
I ask this question because I may do some of the ELISA steps in the scanning facility.

Any step in the ELISA procedure is critical for trustworthy, reproducible results. Transporting a loaded ELISA plate is no good since you might compromise temperature and volume stability (sun exposure, splashing) which can greatly affect your results.
I highly recommend transporting the samples in a cooler container along with the ELISA kit(s) so you can start the ELISA procedure from scratch where you plan to read the plate measures (at the end of the ELISA). That is how I did it when I was in a similar situation as yours.

All the steps in ELISA shoud be incuated at optimum temperature and incubation time to get reliable and reproducible results. The temperature variation during transportation may affect the results.
I did the ELISA where the ELISA reader facility was there. So I recommend you to do the same.
