The non-ionic detergent can be used as additives in the PCR to reduce the secondary structure, which might be helpful for the low yield PCR. But it may also increase non-specific amplification. Can anyone please help explain the effect of the non-ionic detergent on the double strand DNA? Like will it affect the hydrogen bond? what is the principle in reducing the secondary structure?
