Preparation of liposome is carried out by thin film hydration method with cholesterol, chloroform and phosphatidyl choline. During the process of film hydration drug is introduced in to the liposome( drug and water). The picture is attached with this.
Try employing isotonizing agents in the liposomal suspension medium. Precipitation is simply the act of gravity. It can be overcome by making sure your vesicles are suspended properly in the medium.
Check the following:
1- before starting evaporation, the lipophilic composition should be completely soluble in the organic solvent.
2- you use a suitable RPM to make a homogeneous film without aggregations or empty spots.
3- before adding the aqueous phase, the lipid phase should be melted (the water bath temperature needs to be above the transition temperature), we evaporate the organic solvent at a low temperature then raise it to a suitable temperature.
4- the aqueous phase should be clear without any precipitation and wormed to the same temperature as the water bath adjusted.
5- the buffer pH and concentration are suitable with the pKa of the components.
