I incoporated anticancer drugs in Calcium phosphate particles, which is supposed to cure bone cancer. Here I want to test the drug release in vitro at pH 7.4 and 6.5(mimic tumor microenvironment pH). so what buffer I could choose?
I found the buffer in the following link. Can I use this to replace PBS?

I think I would avoid using a phosphate buffer for this experiment because the particles are made of phosphate. Instead, you could use an organic buffer. For pH 7.4, HEPES is a good choice. Tris would also be acceptable. For pH 6.5, I would recommend MOPS.

I have an idea. Purpose of release is to mimic the in vivo situation, so PBS is acceptable for pH7.4 since in vivo environment has PO4-, in this way the HBSS seems a better alternative than PBS as it mimics more (but PBS is also ok, as release curve only give an impression and trend.
But for pH 6.5 is really headache. so I would delete this group, as release in buffer is not cell mediated, which is at least 3 times faster than buffer based on previous result.
so, In the end I will only use PBS 7.4 to have the trend and impression to compare my 3 materials.

As far as I know from biochemistry classes, and knowing the main buffering agent in culture media, the best choice for pHs like, 6.5, is to be bicarbonate which pKa is exactly 6.5. I hope that will be helpful!
