I found bacterial concentration of a culture solution with serial dilution method (e.g. 10^7 cell/ml). Now, I want to transfer a given amount of the solution (e.g. 1ml) to the same but fresh (without bacteria) media in order to inoculate 10^7 bacteria into a new media. But before transfer I need to wash the bacterial solution by adding PBS and centrifuging. Does this washing change the bacterial concentration? Has 1ml of washed solution the same number of bacteria as 1ml initial culture solution?
When you transfer 1.0mL of 10^7 cell/ml into a new medium, the concentration is not the same but the number of bacteria is still the same if you work fast because given a suitable environment for growth, they divide and double in 20min. The question is how fast and efficient is your retrieval. Centrifuging alone could not recover all the bacteria.
Why do you need to wash your bacteria? Will it not suffice to transfer them in new culture or maintain them in a chemostat?
Dear Hamed, I have the same doubt as Buenaflor's. Why do you need to centrifuge and wash your bacteria before transfer? If you are transferring to same media you could just directly inoculate the fresh media with 1 ml of bacterial culture. Centrifugation can damage your bacterial cells and the number of viable cells will not be same. Please go through the article- http://aem.asm.org/content/78/1/120.full

Centrifuging your bacterial cell pellet will lead to disruption of cells. The viable cell count will decrease.
