I isolated and purified plasmid DNA from my E. coli culture using protocol of Addgene.
They say overnight culture at -20 °C would help precipitation of plasmid DNA in case of using 70% alcohol for precipitation.
But I used 2-propanol instead of the alcohol. The technical representative of Addgene says
it might hurt my plasmid DNA.
So I wonder, is overnight precipitation of plasmid DNA in 2-propanol harmful for my DNA??????
I got hard pellet.
The DNA itself certainly does not get hurt. But you precipitate a lot of salt that way, that ends up with your plasmid. Might be OK. Depends on your downstream application.
You could try to give the pellet some extra thorough washes with 70% EtOH (not IPA) to get rid of some of the salt.
