I am using Qiagen blood and tissue kit for extracting of DNA from ticks I elute the DNA after incubation 1minute as manufacturers instructions
If I increase the elusion step to more than 1 minute to maximize the DNA yeild does this decrease purity of DNA?
In my experience, increasing the incubation time over 1 min doesn't increase the DNA yield. However, I've done a single elution, (with 100 uL) for example, and taken that 100 uL and put it back on the column for another minute and gotten higher yields. I didn't elute with a separate 100 uL and combine the two because I didn't want to lower my DNA concentration. Hope that helps.
I believe it increases yield from my experience but not purity. Robert Drake Mitchell III is right about re-eluting for extra yield.
