Coming from a background of materials and X-ray science, I have little knowledge about endocytosis and cytotoxicity. I wonder if I add gold nanoparticles (AuNPs, citrate modified, ~2-5 nm in size) into cell suspensions, and some of the AuNPs happen to reside on top of the cells, is the gravity of AuNPs strong enough to drive them to penetrate the bilayer?

It seems that gravity can play a role, but not in the way I assumed. The authors claim that sedimetation leads to a higher concentration at the bottom, thus more uptake of AuNP (by endocytosis). However, the AuNPs therein are relatively large (>15 nm).
If smaller ones are used, e.g. 2 nm, maybe direct penetration can become significant? BTW, why you think cells will lyse?
