DiBAC4(3) (HY-101892, MCE) is a slow-response membrane potential-sensitive probe. Its working principle is based on the electrochemical response to changes in membrane potential: Upon membrane depolarization, the negatively charged DiBAC4(3) probe enters the cell and binds to intracellular proteins or membrane components, resulting in enhanced fluorescence and a red shift in the emission spectrum. Due to its anionic nature, the transmembrane movement of the probe further amplifies the depolarization effect, forming a positive feedback loop that significantly increases the fluorescence signal. Conversely, a decrease in DiBAC4(3) fluorescence intensity indicates membrane hyperpolarization.
