I am trying to obtain a decellularized pig bladder conserving the basement membrane. However, when doing the staining for Collagen IV and Laminin (with human antibodies), I can see clearly positive blood vessels but a negative urothelial basement membrane surface, both in control and decellularized samples, which leads me to think that the epitopes are different or something is altering the surface during the tissue preparation for IHC but not the blood vessel. So I was wondering if someone else has come across this problem.

Any piece of advice is welcome.

Thanks in advance.
