I was trying to assay the viabilty of my sponge cells after two weeks and the test with DMSO came out blank while the test with conc NaOH gave off Purple colour. Out of curiosity I added MTT without any cell into an ependorf and NaOH and surprisingly the mixture still turned purple. I did the same thing with DMSO and MTT and there was no reaction.

The appearance of the purple coloration is an obvious indication that your MTT dye has been reduced to the purple formazzan form. The obvious question is, can NaOH act as the reducing agent? Yes because in solution it is present as Na ion and hydroxide ion, the hydroxide ion is one common reducing agent that is sure to reduce your MTT.
