I'm preparing a chitosan hydrogel and i want to incoporate PLLA nanoparticles within. To produce the hydrogel, the chitosan is dissolved in acetic acid. PLLA nanoparticles are mixed and further, the solution is reticulated with GPTMS. I need to know if acetic acid can dissolve the PLLA nanoparticles, and if the reticulation agent is influenced by the PLLA presence.
Moreover, in a collagen assay with human gingival fibroblasts and PLLA nanoparticles cultured on top, this was the results i got for PLLA concentration of 1000 micrograms/mL. Looks like sirius red was highly absorbable by PLLA. Does PLLA react with sirius red?
PLLA is a polyester and therefore don't have any functional groups. Therefore Sirius red, acetic acid and GPTMS will not react with PLLA. i don't think PLLA nanoparticles will dissilve in acetic acid at the concentrations used for the dissolution of chitosan.

I cultured PLLA nanoparticles above a cell culture of human gingival fibroblasts. I then performed a collagen test. In higher concentrations of PLLA, it looks like the nanoparticles absorbed most of the Sirius Red - Image in appendix. How is it explained?
