I usually work in the laboratory alone, but sometimes, somebody from Plant Pathology department comes and uses the thermocycler in my lab, yesterday I saw that she turned on the thermocycler and ran the program, after a while she opened the lid of the thermocycler (without stopping the program) and put the PCR microtubes into the wells.
Is it dangerous?
Should I warn her?

In some of the thermal cyclers, an error is generated if the lid is opened before the program is over. Opening the lid during the process may result in undesired results in amplification.
The machine gets sudden pressure due to an unexpected change in the command if the lid is opened.The machine may cope up with it, but the practice of doing so is not recommended. There are options to abort the run. However, if it is done unknowingly for once, its ok.Forgiveness is always the better option!

No, she didn't run a "Hot Start" program.
I think I should warn her, they destroyed two thermocyclers in their department and I think they are going to do the same thing in my lab!!
