I checked the nanodrop conc of the plasmid sample and it was 1000 something. I cleaned the platform and the upper arm got mistakenly lowered and it still measured the conc around 41.Could be due to the leftover sample on the platform that wasn't cleaned properly. However, the ratios remained the same. Does that mean that the nanodrop concentration depend on the volume of the sample taken?
No, it should not have to be.
It's designed so that when you close your lid, and the drop of the sample is dispersed, the width of sample is the same for all measurements.
It should not be volume dependent but very small volumes may evaporate and give a high concentration. it may be that you have old dna bound to the pedestal optical lens . Normally zering (blanking) with your solvent deals with this but it can be cleaned off using 0.5M HCl if water washes are not removing the contamination

The assay is not dependent to the volume of sample you load on the optical lens of Nanodrop, but the lens is tiny and you couldn't cover it with more than a few microliters, and in the other hand the software connected to Nanodrops mostly shows the concentration based on ng/ul.
Mostly wrong concentrations are given due to the dirty lens from last assay. It is important to clean it enough also with a soft cloth or tissue paper to not scratch the lens.
As Paul Rutland also said running blank with the solvent of your nucleic acid (mostly water or TE buffer) is so important. I recommend to first blank with the solvent and then run one time with your solvent again to make sure is cleaned enough, If you wont find any nucleic acid within the solvent then its ready to measure your sample concentration.

The proper optical way (clean your lid with special soft tissue) is a key. Low volume mean absent of clear optical way. Always add blanck before measure with the same solvnt as in your sample.
