Some RIPA buffers have 0.1% SDS and/or 1% Triton-X100 so they solubilize nuclei also, resulting in a highly sticky solution. Just using NP40 or equivalent (deoxycholate or CHAPS) gives better results especially for membrane bound receptors and proteasome preparation.
