Recently, in fluorescence microscopy observations, it was found that cell lines induced by Doxycycline (HY-N0565, MCE) showed blue fluorescence signals. It is not clear whether it is the characteristics of the drug itself or interference from the experimental system (such as fluorescence filter matching problems).

Doxycycline itself is not a strong fluorescent compound, but its molecular structure can show fluorescent properties under certain conditions. Experimental observations show that when doxycycline acts on cells, a weak blue fluorescence signal can be occasionally detected under ultraviolet light excitation (usually excited in the 405 nm range).(PMID: 24287973)
This fluorescence characteristic may interfere with the results of conventional cell experiments, such as interfering with the detection of other fluorescent probes or fluorescent proteins.
If relevant signals are observed in the experiment, it is recommended to combine negative controls (untreated cells) and spectral analysis to eliminate nonspecific interference. Alternatively, fluorescence interference can be reduced by controlling the concentration and usage time.
