I am wondering if the immunofluorescent staining of CD31 antibodies can both mark endothelial cells and indicating their viability. In other words, if I perform immunofluorescent staining for CD31 on a sample of HUVEC, will the signal correlate with the cells' viability?
Your help and relevant references are highly appreciated.

The short answer to your question is - no :(
To my best knowledge and based on my experience CD31can be detected on both live and dead cells, and even on isolated cell membranes. One may find some insignificant changes in CD31 levels in dead cells, but it is not a clear cut between the healthy and injured/dead cells.

Just to clarify: from your question I understand that you are trying to correlate the CD31 levels with cell viability=death (e.g. you expect to see reduced levels of CD31 in dying/injured cells). HUVEC express appreciable levels of CD31, which does not change markedly under various noxious conditions. You may observe re-localization/re-distribution of CD31 at the cel-cell junctions in disturbed/retracted HUVEC monolayers (e.g. you may observe more diffuse CD31 immunostaining). However, the overall levels of CD31 will not be markedly changed. In this regard CD31 is not a good marker of cell "viability=death". Lack of CD31 immunostaining at the cell-cell junctions may only indicate disruption of the HUVEC monolayer integrity, but that doesn't mean that these calls are injured/dying.
Dr. Chernov's referred paper deals with the physiological role and signaling of CD31, and to my best understanding does not address CD31 as a "marker" of cell viability/death.
