I previously labelled neurons with biocytin-containing K-gluconate internal patch clamp solution, and it works great.

However, for a new experiment I am using using a high chloride cesium-based internal solution, and I would also like to fill and label those neurons. I am concerned that using biocytin in this internal might work less well than for K-gluc internal.

For example, one paper advises:

"In our experience, using cesium- and chloride-based internal solutions reduces the ability to recover immunostaining for certain neurochemical markers. Neurobiotin or a cell-impermeant, fixable, polar tracer combining an Alexa Fluorophore with biocytin can be used as an alternative to biocytin."

I was wondering if anyone here has experience using biocytin with a cesium-based internal? Did it work well, and if not, what dye do you specifically recommend instead?

Thank you in advance!
