Generally, the binding of drug with serum albumin can not affect the conformation change. If you want to confirm, then you perform Mass spectroscopy of the conjugate.
Does the conformation change always indicate that the drug is binding to sudlow site I, or binding of drug to any other site can also cause the conformational change.
