I'm working with porcine pancreatic lipase type II, and I know that its specific activity measured by using olive oil (30 min incubation) is 100-400 U/mg protein but I would like to know its specific activity using spectrophotometric assay with p-nitrophenyl laurate
Specific activity is a function of enzyme purity ( how pure is that) and its specificity for a particular substrate. Having said that, the sp.activity purely depends on the way the enzyme is prepared ( how crude or pure it is) and it varies with substrate. The values needs to be established, however, the sp. activity of the pure form of Pancreatic lipase should be available in literature. 30-60 U/mg for triacetin and you need to establish with you substrate if there is not value in literature.
