I performed western blot to detect the "a" determinant region of the HBsAg in my protein sample. This same protein has 6x his-tag. With anti-His antibody, I obtained two bands but with anti-HBsAg, only one of those bands appeared. It makes me wonder why the Anti-HBsAg cant detect both bands.
Also, does anyone know the sequence on the HBsAg that is the epitope to which the monoclonal anti-HBsAg antibody binds?

What is the composition of your protein sample? Is it a purified protein, or a mixture?
Which monoclonal antibody are you using? I know several mAbs recognizing determinant a , but different sequences within this region. You have to search for the particular mAb you are using, perhaps there are epitope mapping studies with it.
