The cells are procured from ATCC and they recommend to use the specified media supplied by ATCC itself. But since sigma is also providing the same RPMI 1640 media, I thought of going with that. But the problem now is that the media composition seems to be varied between sigma and the one recommended by ATCC. Does anyone have a suggestion of which media to use? Does anyone know all components for preparing complete RPMI 1640 media for culturing THP-1 cells?
I have grown them in regular RPMI (glutamine included, from Wisent, Canada) with 10% FCS and I think RPMI from any media supplier should be fine. No other additives are necessary but if they are slow to start up its a good idea to control the pH by adding 20mM HEPES, and provide extra serum initally.
I have used RPMI 1640 + 20 mM HEPES, 0.85g/L NaHCO3, 10% FCS (heat inactivated), 20 mM Glutamine, 1000U/ml Penicillin and 1mg/ml Streptomycin. The ATCC recommend addition of 2-mercaptoethanol to a final concentration of 0.05 mM, I haven't tried this myself, but apparently this could improve the cell growth by preventing the accumulation of toxic levels of oxygen radicals (it is very unstable in solution so it has to be supplemented daily)
How about using RPMI-1640 (from Sigma-Aldrich) supplemented with 2.0g/L NaHCO3, 10% FBS, and 1mM sodium Pyruvate? I have used this components to prepare the media. ATCC recommends to use 2-mercaptoethanol and additional 4.5g/L glucose,which I haven't used (since RPMI 1640 media from Sigma already contains 2g/L glucose). Do I have to heat inactivate FBS? Want few suggestions regarding the same.
