I want to look for galactose-alpha-1,3-galactose in ticks and for the presence of mammalian blood at the same time. I only found primers for vertebrates or species specific ones. But i only need to know wether there is mammalian DNA detectable or not.
you should pick a specie to work with, because what you're asking for is a primer for a conserved region between all mamalians, right? i really don't know either, but in NCBI you can look for an enzime DNA, galactose-alpha-1,3-galactose is a carbohidrate, so you want to do PCR of what gene?

Interesting question!
A quick literature search finds some interesting candidates, including the mammal-specific histone H2A variant H2ABbd (see link below) or the mammal-specific N-terminal extension of the Wilm's Tumor 1 protein (see second link below). You will want to try and design primers for these mammal-specific proteins (or protein extensions, in regards to WT1) that can amplify as many mammalian species as possible, so some bioinformatics work will need to be done.

You can easily check for human vs non-human DNA by using a human specific sequency in the DNA. Alu sequences are present in a lot of mammals, but some recently developed sequences are primate specific or even human specific. AluQuant is a kit that makes use of this and can also quantify the amount of human (and you even have kits that can determine the precense and amount of Y/male DNA) DNA present in your sample. Also the AluYd6 is a primer which can be used for human specificity.
