Many studies have used Caco2 cells for these experiments - however, Caco2 cells are colorectal carcinoma cells, and are not the best model for nutrient transport in the small intestine. I am currently attempting to polarized Hutu80 cells (derived from duodenum), but they do not appear to form tight junctions. Can anyone suggest a cell line that would work?
Most people use CaCo2 for making polarised sheets and measuring transport across these.
During my Phd I used HT29 cl.19A cells which were polarized, and used to study secretion and filtration in cholera disease for example
How about trying a mouse cell line and modelling this alongside the polarised human colorectal cells.
Caco-2 cell lines grown in polycarbonate membrane transwell inserts will differentiate to polarized monolayer after one week culture in DMEM containing 10% (v/v) fetal bovine serum (FBS) high glucose, L-glutamine, pyruvate, and fortified with penicillin, streptomycin and gentamicin . They will mimic small intestinal epithelial cell organization having basal membrane,apical microvilli and well established apical junctions after 7-8 days will be ideal model to study nutritional uptake.
I also used CaCo-2 for polar transport across the cell sheet on Transwell membrane:

You will see pretty nice microvilli on the cell surface by SEM analysis:

Caco cell line is colon cancer line, but these cells can be polarized after about one week on the polycarbonate membrane in transwell. DMEM and 5% FCS should be OK, but without pyruvate.
Hi everyone, thanks for responding. My question was about an intestine-derived cell line that is capable of being polarized. We *do not* want to use Caco2 cells (or HT29) because they are derived from the colon and not the best model for nutrient uptake in the small intestine (i.e. the jejunum). Has anyone heard of or used MKN7 cells?
