I am interested in a particular microRNA produced in the kidney but also found in the circulation. I would like to isolate circulating exosomes and then identify by surface markers those excreted by the kidney. Does anyone know if there is such a marker? I haven't been able to find it. Thanks heaps in advance.
Renal epithelial cells in the proximal tubular lesion, which is the portion of the duct system of the nephron, secrere high amount of exosomes including miRNA. For instance, the proximal-to-distal vesicular inter-nephron transfer is well-known. CD63-positive exosomes secretion is promoted by angiotensin-II, which is activated by ACE (angiotensin-converting enzyme predominantly which exists in the pulmonary epithelial cells). In the perspective of the function of miRNA in the "capsule" exosomes, microRNA-29c in urinary has been identified as a novel biomarker of renal fibrosis.
Importantly, urinary miRNA does not limit to the kidney disorder, as you know well. For instance, exosomal miR-21 expression is up-regulated in serum from patients with esophageal squamous cell cancer as compared with serum from patients who have benign diseases and was positively correlated with tumor progression and aggressiveness.
