I am studying the the interactions of DMPC and viral peptides using SAXS measurements. The problem is that our scattering pattern for DMPC unilamellar vesicles is weird, the second lobe is flat. This is different to all the other PC-head lipids that we found in literature. We tried several methods for preparation of the vesicles and we cannot avoid this strange pattern. Does anyone know how to avoid this? Or What is the meaning of this flat lobe?

are you able to post an image so we can see what you mean in this case? It sounds like the vesicles are not forming. Do you also have access to DLS to check that you are making vesicles and it is not just a problem with the scattering experiment?

It depends. you should load a 2D pattern (or the scattering profile). this is clearly due to the interaction between vesicles and peptides. I am not sure if there are models but of course raw data are needed.
do you homogenize in size your vesicles by means of e.g. an extruder?

I understand you obtain a strange DMPC-peptide pattern in comparison with DMPC alone. Clearly, it's due to the lipid-peptide interaction. Have you calculate the electron density of layers? Have you checked the morphology by electron microscopy? or Differential scanning calorimetry?
