I want to measure leaf vein density : vein length/ leaf area. I am struggling with ImageJ to obtain leaf vein length. Does anyone know how to do this?
Set the scale, draw a line along the vein and use Analize>Measure tool. It's simple. Here some tutorials:

thank you. I had already tried this but as you can see from the attached image the architecture of leaf venation is quite complex. Using the freehand line is not very practical.... Would you suggest the analyse particles tool?
You can try to do it with LEAF-GUI.

I just work on similar thing. I need to measure vein lenght of alfalfa and I'm too struggling with it. Would you advise me, how did you do it eventually? I'm starting to be desperate. Did you use the Leaf GUI? Or stayed on ImageJ?

I am not working on this anymore but a colleague of mine told me that they are very happy with this :

They take pictures of cleared leaves with dark field microscopy. An option to increase the contrast is consider leaf vein staining.
