Does anyone know how to isolate cells from a 3D matrigel matrix for protein analysis (e.g WB), without getting the proteins of the matrigel?

I already tried with Trypsin/EDTA and Dispase but i still have a lot of the gel in my cell pelet (also after lots of washing steps). Is there a clean way to isolate the cells from the matrigel for westernblot analysisn without having the proteins of my matrigel in the cell lysate?

HI Meltern,

you should use collagenase and dispase in order to digest completelly the matrigel, this shoud avoid matrix fibers contamination in the cell pellet!!!!

Could help the paper: Scchetti, Funari et al., 2007.
