I am looking for a protocol that allow to reproducibly perform LTT to diagnose idiosyncratic drug reactions with SOPs to define drug concentrations and time of incubation (and if possible, cut-offs)
See an article we wrote in the 1960's: Coulson and Chalmers, 1966, Quantitation of Peripheral Blood Lymphocyte Cultures, Nature, 209, 378. This method involves physically counting the actual number of small lymphocytes that have transformed; yielding an index of response.
This addresses the time factor (90 hours of culture), the viabilty of the cells, and the 'background" spontaneous transformation of cells.
Manually counting the cells is the 'best' way in my opinion, but is time consuming. So many researchers use indirect methods such as DNA synthesis.
