I have a protein which has phosphorylated and un-phosphorylated forms. Phosphorylated form is in the cytoplasm and unphosphorylated form is in the nucleus. On my western blots, I see the phosphorylated band, but un-phosphorylated band is either weak or absent. So, I think I might not getting enough protein out of nucleus.
Any suggestion about nuclear protein extraction or western blot for phosphorylated proteins is appreciated.

Nowadays there are quite a number of kits available in the market that can be used efficiently to extract nuclear specific proteins. NE-PER Nuclear and Cytoplasmic Extraction Reagents is a good one. I would suggest you to buy this product. This works fine.
