I have tested some chemicals on lung cancer cell lines by using WST assay with 24 hours incubation time and finally I got the result that these chemical gained higher cell viability when compared to the control. How can I explain this event?
If your plate layout is somehow correlated with your results, you should check for a systematic error. Generally, when you repeat your experiment (to check for reproducibility), you should change the layout, e. g. a compound that was tested in plate column 12 should then be tested in plate column 3 (assuming columns 1 and 2 are used for controls).
Did you test your compound without cells? It might be, that your compound reacts with the WST itself. You might have this effect with cigarette smoke for example.
