My samples are groundwaters (20-80 m deep). I will analyze the sterols using HPLC-UV and GC-MS. We have acquired a HS-SPME autosampler and I want to run some tests with those substances and if someone can give me a starting path to follow, it will appreciate it.
You may like to check out Parkinson et al. (2010). Different matrix but it should give you an idea about the derivatization step for your rather non-volatile sterols for GC-MS and a suitable choice of fiber.

I would prefer a liquid-liquid or a SPE extraction followed by a derivatization with TSIM. HPLC-UV is not a good technique for analysing neutral sterols.

Chistoph, I also think that HPLC-UV is not the best technique for sterol analysis. Fortunately, our GC has just arrived. We are moving to most environmentally friendly techniques, such as SPME, but we know that not always is possible its use.
