I assume you are interesting in orthotopic lung cancer mice models. For that case there are basically different approaches: i) Percutaneous injection -usually blinded under fluoroscope monitoring, with high rates of leakage and pneumothorax; ii) Surgical orthotopic implantation -via thoracotomy, very precise delivery of the implantation but technically challenging and associated with high post-surgical mortality; iii) Transpleural orthotopic injection -minimally invasive procedure, blinded but with very precise implantation with low morbi-mortality. You can find an interesting paper that compare these 3 techniques and their outcomes by bioluminescence monitoring. They also include a brief description of the methods for orthotopic implantation.
