I plan to amplify environmental prokaryote from sediment or water. Therefore I searched for a generalist primer set targeting a short region (max size of amplicon=500pb). I think that such primer exists for 16S gene. However I have no experience on this group. Can you help me?

My project is now at the beginning, just an idea. For now I try to assess its feasibility. This project will be based on my ability to amplify Prokaryota DNA present in the river. Although my lab has good experience on molecular ecology, nobody has ever work on prokaryota.
So I need to get feedbacks from researchers that work on prokaryota.
Concretely I search for universal PCR primers that target a large diversity of procaryota found in freshwater environment, as we can found universal 18SrDNA PCR primer targeting eukaryota. Is someone have such primer in his bag? It sounds too ambitious?
Usually 27f & 518r for bacteria and 21f and 518r for archaea. Dr. Nunoura published primer set common for 2 domain. Please check it.
