I tried to incubate 2mM glutathione with late-log L. amazonensis promastigotes for 16 hrs and the leishmania was dead. I am not sure whether GSH kills leishmania or the GSH has been converted to anything that kills parasites. Previous literature had used this concentration to treat L. amazonensis. It seems so strange that GSH will kill leishmania.

GSH is not directly going to reduce the oxidative stress developed inside the parasites. There is some alternative pathways are existing. I'm not sure, that may be trypanothione pathway. When you use some of the intermediate in that pathway you may get recovery.
Moreover I feel you have used very high amount of GSH. Usually micromoles of GSH or less GSH present in our cells. In human, side effects of GSH is not reported. So in parasites we can't correlate the GSH level with toxicity. Verify the information what you have and then perform the experiment.
