We have some doubts in differentiating with other structures and we want to know how to confirm if biofilm in tissues (attached photos, SEM, lung with culture of H. somni).
I attended yesterday a talk given by Prof. Dr. Oliver Lieleg from Technical University of Munich. He is working with biofilms and I think that his research may help you to clarify your question.
The difficulty with using SEM is that the fixation and sample preparation can sometimes remove the "characteristic" matrix of biofilms.
Another difficulty is that in vivo biofilms are often not what expect to see based on in vitro work, for instance this matrix may not even be present in in vivo samples.
I have attached two papers you may find helpful, hope this is useful.
In most of the works that identify biofilm in tissues with chronic infections, they perform H & E, PAS and FISH. The SEM seems complementary. Thanks for your contributions!
