This kit (https://altogen.com/product/mrc5-transfection-reagent-diploid-fibroblast-cells/) has a designated CRISPR/Cas9 transfection protocol specific to MRC-5. I would also look at previous studies to see if anyone else has implemented CRISPR in MRC-5 (it might be difficult given that it is a fibroblast cell line) and see what methods they used as well.
