I'm recently working on mouse tendon cryosection for immunostaining. But when I cut these tendon samples, they scattered so it's very hard to get intact sections. I've tried to changed the cryostat temperature up or down, and also tried snap freezing fresh samples or 4% PFA fixed and 30% dehydrated samples. Does anyone have experience with tendon cryosection?

Thanks Mohamed. How long do you fix the tendons before dehydration in sucrose? I did it for 24h. Another thing is when I cut the sections off and attached them to slids, those sections can be attached completely. Looks like the tendon sections are wavy and some parts of it can't be attached to slides. Did you see the same thing when you worked on tendon sectioning?

Have you tried to adjust the temperature in the cryostat? -30 and more?
Sometimes thinner sections have less chatter than thicker sections; try sectioning at 8 microns. Also, have you explored any tape transfer techniques?
