My work is to attenuate resistance gene which is adenine autotroph and i want to confirm mutant clone on M9 minimal media .recently I use 5 x M9 with 1 M glucose and adenine but it did not grow in M9 + glucose even wild type bcc strain . Does anybody know how much concentrations of M9 salt suitable for adenine auxotroph and how to prepare?
