If you intend to do this using a chemical electrophile, it would depend on which nucleophilic position in the purine ring you wish to add the methyl group to. For example dimethyl-sulfate will add a methyl group to the N-7 position.
Look up the very old chemical DNA sequencing methods (Maxam and Gilbert). The DNA backbone is more prone to cleavage at sites were the adenine is methylated under certain conditions. This action was used to in the 1970s to sequence DNA.
