I am working with an unstable plasmid that mutations occurred during cloning in JM109. I'm wondering if any of you have the similar problem and how you work around this. Thanks for the help!
Yes, unstable, toxic or repetitive genes can be a serious pain. I suggest growing the plates and the liquid cultures at 30C instead of 37C and to use richer media (e.g., super medium) and not plain LB. As for strains with higher stability, I had a good experience with Stbl3 by Life Technologies, but even when using them I also used rich media and 30C.
