We plan to sequence several samples isolated from rat brain hippocampus. My previous colleagues used NextFlex version 3, which was working fine, and now we are continuing with NextFlex v4. The difference between these two kits is especially in size selection and the fact that NF v4 is gel-free in comparison to v3. Unfortunately, we are getting low yields of the library - miRNA control provided by the manufacturer is 1 ng and our samples are around 0.2 ng after preparation. We hope it's not a matter of the kit functionality but rather something we can optimise.
